Inference of Cell Fate Reprogramming

Network medicine is an emerging field characterizing the disease as the result of perturbations on molecular network. Thus, the transition between a healthy and a diseased state cell or conversely is interpreted as a reprogramming induced by the topological perturbations of the molecular networks that leads to alter the cell fate. The challenge is to infer these topological actions characterizing the causes of the cell fate change. We propose a theoretical framework, called Boolean control network, modeling the control resulting of the application of the topological actions in the dynamics of network. The inference is based on abductive principle deducing from the effects, the disease or the treatment, the parsimonious topological actions assimilated to the causes. The modeling framework can be applied to a large number of biological situations related to the cell fate alteration. In the talk we discuss the fundamental aspects of this framework and their application to complex diseases.
